CD80 is a costimulatory molecule that serves as the ligand for two molecules expressed on T lymphocytes, CD28 and CTLA-4. The cell-surface molecule is found only on antigen-presenting cells. It mediates critical T-cell inhibitory signals. Like CD86, CD80 is a type I glycoprotein made up of two extracellular lg-like domains, a transmembrane region and a cytoplasmic tail. The N-terminal V-domain and the membrane-proximal C-domain share high homology to each other and the respective regions of lg’s.
The B7/CD28 CTLA-4 costimulatory pathway has been shown to play a critical role in determining the fate of immune responses, meaning activation versus down-regulation, and is a highly promising therapeutic target for treating autoimmune diseases. The regulatory roles of CD80 in autoimmune pathogenesis have been shown for a number of diseases like systemic lupus erythematosus and multiple sclerosis. It is furthermore a crucial molecule in transplantation biology.
