AHP1233 recognises glycogen synthase 3 (GSK3) beta phosphorylated at serine 9. This is a 46 kDa member of the serine/threonine protein kinase family. Two isforms of GSK3 exist, alpha and beta. GSK3 beta is expressed in the testis, thymus, prostate, and ovary as well as being expressed at low levels in lung, brain and kidney tissue.
GSK3 beta is involved in many signalling pathways including energy metabolism, body pattern formation and microtubule growth and stability (which aids axon growth). GSK3 beta is also thought to be involved in oncogenesis and the hormonal control of several regulatory proteins including glycogen synthase.
Phosphorylation of serine-9 inhibits GSK3 beta function.

For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
